| Resumen: | The scope of this work item is to develop a new standard for determination of total lipids in micro- and macro algae. Current methods applied for other types of samples lead to an underestimation of the total amount of lipids, since membrane phospholipids and glycolipids are not extracted by current methods. The content of these lipids can amount up to about 40% in algae, while in normal organic material (like plants) this only reflects 1% of the total lipids. This can lead to a considerable underestimation of the total amount of lipids in the algae. Good experiences are obtained with the help of the Ryckebosch-Foubert method. This method makes use of an extraction protocol with chloroform-methanol 1:1 as a solvent. Therefore, the Ryckebosch-Foubert method will be used as starting protocol for the development of the new standard. It needs to be stressed that this proposal concerns a lab method to determine the total amount of lipids. It is not the aim to have this method (using chloroform) adopted for large scale commercial extraction, for which other solvents should be used. Yet in order to optimize these commercial solvents, it is important to know what the total amount of lipids present is, so that the extraction efficiency (amount extracted with commercial solvent / total lipid content) can be calculated. |
